Applications of BBa_K2073010
Firstly, we aimed to know the effect of fluoride on E.coli. We cultured E.coli in LB with different sodium fluoride concentration and traced the growth curve of E.coli in fifteen hours.
The result shows that fluoride significantly inhibits the growth of E.coli. The higher fluoride concentration , the less E.coli grow.
In the LB with 0.1M sodium fluoride, the E.coli stops growing.
Secondly, we tested the protein function of Crcb-flc. After transfecting Crcb-flc gene into E.coli, we cultured them in the LB with 0.1M sodium fluoride.
The positive control is E.coli cultured in LB without sodium fluoride.
The negative control is E.coli without Crcb-flc gene cultured in LB with 0.1M sodium fluoride
Based on the result, in the LB with 0.1M sodium fluoride, the E.coli with Crcb-flc gene can still grow. (Crcb-flc-test 1,2)
However, as for the E.coli without Crcb-flc gene, the growth is completely inhibited.(negative control)
We can know the Crcb-flc successfully exfluxes the fluoride and promotes the growth of E.coli.
